The analysis of humic compounds by elemental analysis, PY-GC-MS and HPLC techniques permitted determination of the origin of organic inputs, essentially terrestrial, and the effects of environmental conditions in the deposits along a north-south transect through the Rhone delta, from the river mouth to the open sea. In the prodelta the sedimentation of fine particles enriches the sediments in organic matter. The humic compound contents were relatively low in sediments, perhaps due to a rapid burying of organic matter which leads to anoxic conditions. Samples of phytoplankton cultures and of suspended particulate matter from continental, brackish and marine waters were examined by HPLC for the monosaccharide content of their hydrolysable sugar fraction. Eight monosaccharides were currently assayed and their weight compared to HPLC-assayed chlorophyll a concentrations. With the exception of oligotrophic waters, these two variables are well correlated in the various prospected environments. Glucose was always the strongly dominant form, not only in fresh and brackish waters but also in marine samples with high phytoplanktonic biomass. Chloropigments and carotenoids were measured by HPLC in an intertidal muddy sediment of Marennes-Oleron Bay (France). Concentrations were determined as a function of sediment depth at low tide. The analyses were carried out at monthly intervals over 1 yr. Pigment analysis indicated that the microphytobenthic community was dominated by diatoms throughout the sampling year. Chlorophyll b was not encountered at any time or depth, indicating that no input from macrophytic detritus had occurred at the sampling site. A HPLC protocol is described which permits analysis of the pigment composition of coastal and intertidal sediments. The HPLC technique has been extensively applied to phytoplankton pigments for fifteen years. Papers dealing with living benthic microphytic populations have been more rare, due to complexity existing in the sediment surface, where pigments (chlorophylls and carotenoids) from several taxonomic groups of microalgae are mixed with degraded pigments from senescent phytoplankton cells and macrophytic material. The egestion of particulate material as well as pigment degradation during microzooplankton grazing on phytoplankton are poorly known processes. In an attempt to evaluate these processes, changes in pigment concentrations within various size fractions were monitored in batch cultures of an assemblage of a pelagic ciliate (Strombidium sulcatum) and a heterotrophic flagellate (Paraphysomonas sp.) feeding on a cyanobacterium (Synechococcus sp.) over a 10-day period. The effects of genetics and environmental factors on isoflavone content of soybean (Glycine max L.) cultivars grown in different locations in Brazil in 1993/94 were evaluated. Seeds of different cultivars were analised by high performance liquid chromatography (HPLC). In Rio Grande do Sul (RS), Paraná (PR), and Mato Grosso do Sul (MS) States, a significant difference in the isoflavone total content average of the cultivars IAS 5 and FT-Abyara (163.9, 116.4 and 79.5 mg/100 g, respectively) was observed. In general, IAS 5 contained higher isoflavone than FT-Abyara. Within the framework of the multi-disciplinary program Seine Aval, phenolic compounds obtained by CuO oxidation have been studied since 1995. Quantitative analyses have shown a terrestrial contribution in flood, characterized by compounds of the group P (polyols + phenol), S (syringic) and H (hydroxybenzoic). During the low water period, phenolic compounds allow a clear distinction between fluvial and estuarine suspended material. Fluvial run-off contained Syringic, Hydroxybenzoic and Vanillic phenols. In contrast, upstream penetrating marine and estuarine waters are characterized by cinnamic phenols originating from estuarine herbus. In the turbidity maximum zone, aerobic degradation of lignin material increases the (vanillic acid/vanillin) ratio. |

The high pressure liquid chromatography (HPLC) was used for the identification of isoniazid (isonicotinic acid hydrazide) in the milk of cattle treated with a dose of 25 mg/kg/day in alternated days. The effect of milk pasteurization on the isoniazid residue concentration was also studied. The drug excretion presented a cyclic variation, with higher levels in the first day after administration (aa), a mean of 1104.48µg/l, and a decrease two days aa, with a mean of 104.12µg/l. Four days after the last administration of the drug it was not possible to identify residues of isoniazid in the milk of treated animals. In order to evaluate the influence of photoperiod on the accumulation of allantoin in comfrey (Symphytum officinale L.), a well known medicinal plant, an experiment was conducted during July and August, 1998. Cuttings obtained from 90 day old plants were submitted to four photoperiods (8, 12, 16 and 20 hours). After 60 days, allantoin content in roots and rhizomes was evaluated. The results showed that increases in photoperiod promoted an increment in the average content of allantoin in roots (0.06%, 0.303%, 1.213% and 4.78%). O objetivo deste trabalho foi identificar e quantificar os açúcares em frutos de pessegueiro da variedade 'Biuti', armazenados sob condições de ambiente (27,3ºC; 70% UR) e sob refrigeração (4ºC; 90%), comparar as diferenças entre os teores de açúcares nas duas condições de armazenamento. A identificação e a quantificação precisa dos açúcares (glicose, frutose e sacarose) foi realizada por cromatografia em fase líquida HPLC. The stability of orange juice obtained from a small extractor and stored in a polyethylene bottle was assessed under isothermal and non-isothermal storage conditions at 4, 8 and 12ºC for 72 hours. pH, titratable acidity and Brix did not alter significantly during the 72 hours storage. Microbiological analysis showed high initial count for moulds and yeasts that increased in the juice stored for 72h under the non-isothermal conditions with temperature abuse (12ºC/4h). In this work, polyclonal antibodies anti-human Factor IX were produced in New Zealand rabbits by immunization with commercial pure human FIX (hFIX) (Octanyne®, Octapharma, USA). The serum containing immunoglobulins anti-hFIX was useful to detect hFIX antigen in human plasma fractions submitted to anionic exchange chromatographic process and with a large yield. Immunoassays (ELISA) using bovine serum albumin, trypsin and peptides generated by cleavage assays with trypsin as digestion enzyme was performed and revealed adequate specificity of the polyclonal antibodies produced. |
